You can set spot colours in Illustrator easily.
Just create the colour you want in the colour mixer then drop it to the swatches pallet. Next double click on the swatch and dialog will open. Give the colour a name and set to spot colour (not process). That's it.
This will make anything that you colour with that swatch print out on a different separation.
If you don't know what a separation is you probably want to disregard this post (or ask).
